In a common coupled method, the residual amounts of ATP are converted to AMP using adenyl cyclase. The ADP turns into ATP which is detected through luciferase. Another method uses at least two steps in the process of peroxide-based detection to create hydrogen peroxide. It is then able to react to Amplex Red which will produce the fluorescent product in the third step. These additional steps are a lot of work and unneeded if you choose a direct format instead.
